چکیده

Osmoregulation is important to living organisms for survival in responding to environmental changes ofwater and ionic strength. We demonstrated here for the first time that exposure of HeLa cells to a hypotonic medium (30% growth medium and 700% water) prominently induced the binding activity of the heat shock transcription factor (HSF). Pretreatment of cells with cycloheximide did not inhibit the induction of HSF-binding activity, indicating that the mechanisms of induction are independent of new protein synthesis. The magnitude of hypoosmotic stress-induced HSF-binding activity was comparable with that induced by heat shock. The induction, as monitored by gel-mobility-shift assay, occurred within 5 min of hypo-osmotic stress and persisted at least up to 4 h in HeLa cells under the hypotonic conditions. The heat-shock transcription factor HSF1 is rapidly activated by either hyper- or hypo-osmotic stress in mammalian cells.

Osmoregulation, the cellular response to environmental changes of osmolarity and ionic strength, is important for the survival of living organisms. We have demonstrated previously that an exposure of mammalian cells to hypo-osmotic stress, either in growth medium (30% growth medium and 70% water) or in binary solution containing sorbitol and water, prominently induced the DNA-binding activity o...

In yeast, loss of Hog1 leads to osmosensitivity of autophagy.

In mammalian liver, proteolysis is regulated by the cellular hydration state in a microtubule- and p38(MAPK) (p38 mitogen-activated protein kinase)-dependent fashion. Osmosensing in liver cells towards proteolysis is achieved by activation of integrin receptors. The yeast orthologue of p38(MAPK) is Hog1 (high-osmolarity glycerol 1), which is involved in the hyperosmotic-response pathway. Since ...

Activation of heat shock factor 1 by hyperosmotic or hypo-osmotic stress is drastically attenuated in normal human fibroblasts during senescence.

We have previously reported that osmotic stress prominently induces the DNA binding activity of the heat shock transcription factor 1 (HSF1). In the present study, we examined the effects of medium osmolarity on both the activation of HSF1 and the programmed cell death in normal human fibroblasts during cellular senescence. The activation of HSF1 occurred rapidly in presenescent (early passage)...

The ionic basis of the hypo-osmotic depolarization in neurons from the opisthobranch mollusc Elysia chlorotica.

The resting potential of identified cells (Parker cells) in the abdominal ganglion of Elysia chlorotica (Gould) depolarizes by about 30 mV in response to a 50% reduction in osmolality and returns to the original potential in 20 min. Cell volume recovery requires approximately 2 h. Thus, recovery of the resting potential is not dependent on recovery of cell volume. The hypo-osmotic depolarizatio...

Pregnancy Outcome with Intrauterine Insemination in Patients with Unexplained Recurrent Abortion Whose Partners Have Abnormal Hypo-Osmotic Swelling Test

Background: Recurrent abortion is defined as three or more consecutive pregnancy losses. We aimed to determine the relationship between male sperm parameters and hypo-osmotic swelling score and recurrent abortion. We also studied whether washing of spermatozoa and intrauterine insemination in patients with recurrent miscarriage and low score hypo-osmotic swelling test in male partners could imp...
